Abstract

Tumor necrosis factor α (TNF-α) and interleukin-6 (IL-6) are key inflammatory cytokines that participate in multiple immune regulation and pathological processes. Fusion proteins constructed by linking these two cytokines can exert synergistic biological effects and have important research value in immunotherapy and inflammatory disease research. In this study, the target gene fragments encoding TNF-α and IL-6 were amplified by PCR technology. The two gene sequences were connected by a flexible linker sequence, and the fusion gene fragment was inserted into the prokaryotic expression plasmid to construct the recombinant cloning vector. After restriction enzyme digestion identification and sequencing verification, the successful construction of the TNF-α/IL-6 fusion protein gene clone was confirmed. This work lays a foundation for subsequent protein expression, purification and functional activity detection, and provides experimental materials for further exploring the joint regulatory mechanism of TNF-α and IL-6 in immune response.
